<p class="first" style="font-size: 12px; font-family: 'Lucida Grande'; color: #333333; text-align: left; margin: 10px 0;">An article this month in <a style="font-size: 13px; font-family: Georgia; color: #2679b9; border: none; font-style: italic;" href="http://www.ncbi.nlm.nih.gov/pubmed/20398927"> Biomaterials</a> illustrates how cells can be grown on nanoporous<br />
UltraSM® membranes. From the abstract:</p>
<p>&#8220;The current study investigates the feasibility of pnc-Si as a cell culture substrate by measuring cell adhesion, morphology,<br />
growth and viability on pnc-Si compared to conventional culture substrates. Results for immortalized fibroblasts and primary vascular endothelial cells are highly<br />
similar on pnc-Si, polystyrene and glass. Significantly, pnc-Si dissolves in cell culture media over several days without cytotoxic effects and stability is tunable<br />
by modifying the density of a superficial oxide. The results establish pnc-Si as a viable substrate for cell culture and a degradable biomaterial. Pnc-Si membranes<br />
should find use in the study of molecular transport through cell monolayers, in studies of cell-cell communication, and as biodegradable scaffolds for three-dimensional<br />
tissue constructs.&#8221;</p>

The UltraSM&reg; silicon membrane provides a first-in-class combination of imaging and cell growth abilities for optical and electron microscopy.<br />

<p class="first" style="font-size: 12px; font-family: 'Lucida Grande'; color: #333333; text-align: left; margin: 10px 0;">Cell adhesion to, and observed growth rates on, UltraSM&reg; silicon membranes are equivalent to conventional tissue culture<br />
plastic or glass cover slips. The silicon composition of UltraSM&reg; membranes is biocompatible, unlike carbon films on cytotoxic, metallic<br />
grids. The native silicon oxide film on the membrane contributes a slight negative charge and a glass-like, cell-friendly surface for<br />
attachment and growth. </p>
<p>The nanometer thickness of the UltraSM&reg; silicon membrane is thin enough to allow light and<br />
electrons to pass without significant diffraction. The millions of 10-50 nm diameter nanopores within this membrane create a highly<br />
permeable film which allows cells grown on either side of the membrane to communicate with diffusible signaling molecules. </p>
<p>For multiple cell-type culture models, the UltraSM&reg; membrane enables co-culture of multiple cell types in a setting<br />
where the cells can communicate with one another by short-range signals and be maintained at physiologically meaningful distances across<br />
the membrane. Combined with the ability to image and analyze both cells on either side of the membrane, the UltraSM&reg; membrane is an ideal<br />
substrate for many co-culture studies.</p>

cell culture are too thick to allow detailed resolution of intracellular structures and contribute significant background caused by membrane<br />
auto-fluorescence. The silicon composition of UltraSM&reg;<br />
membranes eliminates concerns over auto-fluorescence, permitting fluorescent imaging at near-UV excitation and emission.</p>

more intense beam currents.</p>

<p class="first" style="font-size: 12px; font-family: 'Lucida Grande'; color: rgb(51, 51, 51); text-align: left; margin: 10px 0pt;">A recent article in <a href="http://pubs.acs.org/doi/abs/10.1021/nl904254g" style="font-size: 13px; font-family: Georgia; color: rgb(38, 121, 185); border: medium none; font-style: italic;"> Nano Letters</a> (N. de Jonge, W. C. Bigelow, and G. M. Veith, Nano Lett., Articles ASAP, Feb. 10, 2010) illustrates how<br />
relatively simple laboratory materials can be used for significant in situ STEM (scanning transmission electron microscope) experiments.  In this paper,<br />
the authors demonstrate the imaging of Au nanoparticles on a thin TiO2 surface in a 0.36 mm thick atmospheric pressure flow cell with a resolution of ~0.4 nm.<br />
The flow cell was constructed with two SiN grids with custom geometry, separated with simple tubing spacers and sealed with vacuum epoxy.<br />
Thin input and output tubing was fed through the specimen rod to the exterior of the STEM for source gas connections.<br />
High resolution imaging through the atmospheric pressure chamber was achieved by focusing the STEM on nanoparticles on the entrance window<br />
(relative to the electron beam), and imaging elastically scattered electrons with an ADF (annular dark-field) detector.</p>

<p>Human neutrophils were placed on IL-8 coated pure silicon TEM window grids (15 nm thick) and allowed to activate. Cells were fixed with glutaraldehyde, then dried with increasing concentrations of ethanol.</p>

These are higher resolution images of the development of the cell&#8217;s leading edge or lamellipodium. Since the pure silicon window was just 15 nm thick, it was not necessary to stain the cells with an electron dense material. Additionally, the optical transparency of these silicon films enables straightforward fluorescence imaging on an optical microscope.</p>

<p class="first" style="font-size: 12px; margin: 10px 0; font-family: 'Lucida Grande'; color: #333333; text-align: left;">The December issue of &#8220;Nature Nanotechnology&#8221; recognizes the 50th anniversary of Richard Feynman’s famous “There’s Plenty<br />
of Room at the Bottom” talk at Cal Tech, which many credit as the first call for the development of a nanoscience and technology field.<br />
Whether or not you share this viewpoint, it’s fun to look back at such a candid discussion of the future by a leading scientist, and to see<br />
how far we have come since 1959.  Feynman was a firm believer in the need to “see” what’s going on at the nanoscale, if we are to truly<br />
understand atomic physics, chemistry, and biochemistry, so it’s not surprising that a significant portion of his talk was devoted to<br />
electron microscopy.</p>
<p>In his feature article, “Surely you&#8217;re happy, Mr. Feynman!”, <a style="font-size: 13px; font-family: Georgia; border: none; color: #2679b9; font-style: italic;" href="http://www.nature.com/nnano/journal/v4/n12/abs/nnano.2009.360.html">[Nature Nanotechnology 4, 786 (2009).]</a> Michael Segal<br />
takes a look Feynman’s predictions and challenges for electron microscopy, and what has actually been accomplished.  Although EM has<br />
not solved all the mysteries that Feynman envisioned, it has come a long way, achieving a 20-fold increase in resolution since his<br />
lecture was given and has uncovered many details of the nanoworld that we take for granted today.</p>
<p>Few of us take full advantage of the power and innovation of modern TEMs, often limited by sample preparation, beam-induced damage, or<br />
support materials.  Next time you are in front of a $1M TEM, spending an hour or more of your time trying to get that perfect image, we<br />
