Since SEM image preparation using Pure Silicon TEM Windows did not require gold sputtering (see our previous blog post), it was straightforward to image the same samples in the TEM. These are higher resolution images of the development of the cell’s leading edge or lamellipodium. Since the pure silicon window was just 15 nm thick, it was not necessary to stain the cells with an electron dense material. Additionally, the optical transparency of these silicon films enables straightforward fluorescence imaging on an optical microscope.
